Once the vibrio cholerae have reproduced and greatly outnumbered non vibrio bacterium a sample can be taken and placed in Thiosulphate Citrate Bile Sats Sucrose Agar. Inoculate for 18-24 hours. After this time yellow colonies should appear of about 2-4 mm in diameter (the yellow colour is due to the fermentation of sucrose in the medium. The inoculation should occur under an optimum temperature with oxygen present (as aerobic respiration is more efficient and preferred by facultative anaerobes.)
